Wind turbine blade failure is the most prominent and common type of damage occurring in operating wind turbine systems. Conventional nondestructive testing systems are not available for in situ wind turbine blades. We propose a portable long distance ultrasonic propagation imaging (LUPI) system that uses a laser beam targeting and scanning system to excite, from a long distance, acoustic emission sensors installed in the blade. An examination of the beam collimation effect using geometric parameters of a commercial 2 MW wind turbine provided Lamb wave amplitude increases of 41.5 and 23.1 dB at a distance of 40 m for symmetrical and asymmetrical modes, respectively, in a 2 mm-thick stainless steel plate. With this improvement in signal-to-noise ratio, a feasibility study of damage detection was conducted with a 5 mm-thick composite leading edge specimen. To develop a reliable damage evaluation system, the excitation/sensing technology and the associated damage visualization algorithm are equally important. Hence, our results provide a new platform based on anomalous wave propagation imaging (AWPI) methods with adjacent wave subtraction, reference wave subtraction, reference image subtraction, and the variable time window amplitude mapping method. The advantages and disadvantages of AWPI algorithms are reported in terms of reference data requirements, signal-to-noise ratios, and damage evaluation accuracy. The compactness and portability of the proposed UPI system are also important for in-field applications at wind farms. 相似文献

Benzo(a)pyrene is a chemical produced during the process of making fried, roasted, and smoked foods. It remains unclear whether benzo(a)pyrene affects the early development of human organs. In this study, we used the transgenic zebrafish line Tg(wt1b:GFP) as a model to assess benzo(a)pyrene-induced kidney malformation. By soaking zebrafish embryos in benzo(a)pyrene at various doses (2, 20, and 200 ppb), only a minor effect on the survival rate was detected (0 ppb: 97.8 ± 1.9 %; 2–200 ppb: 89.1 ± 5.8–91.5 ± 8.3 %). However, benzo(a)pyrene significantly affected the development of the kidney (malformation rates ranges from 50.0 ± 3.5 to 77.4 ± 5.3 %). Various abnormalities, such as unusual curving of pronephric tubes, swollen glomerulus, and incomplete development of pronephric ducts, were observed. This study provides a rapid and effective protocol for the evaluation of the notable effects of benzo(a)pyrene on embryonic kidney development. 相似文献

We examined the acid–base properties of water films adsorbed onto a Ru(0001) substrate by using surface spectroscopic methods in vacuum environments. Ammonia adsorption experiments combined with low‐energy sputtering (LES), reactive ion scattering (RIS), reflection–absorption infrared spectroscopy (RAIRS) and temperature‐programmed desorption (TPD) measurements showed that the adsorbed water is acidic enough to transfer protons to ammonia. Only the water molecules in an intact water monolayer and water clusters larger than the hexamer exhibit such acidity, whereas small clusters, a thick ice film or a partially dissociated water monolayer that contains OH, H2O and H species are not acidic. The observations indicate the orientation‐specific acidity of adsorbed water. The acidity stems from water molecules with H‐down adsorption geometry present in the monolayer. However, the dissociation of water into H and OH on the surface does not promote but rather suppresses the proton transfer to ammonia. 相似文献

Anti‐MUC1 monoclonal antibodies (mAbs) are powerful tools that can be used to recognize cancer‐related MUC1 molecules, the O‐glycosylation status of which is believed to affect binding affinity. We demonstrate the feasibility of using a rapid screening methodology to elucidate those effects. The approach involves i) “one‐bead‐one‐compound”‐based preparation of bilayer resins carrying glycopeptides on the shell and mass‐tag tripeptides coding O‐glycan patterns in the core, ii) on‐resin screening with an anti‐MUC1 mAb, iii) separating positive resins by utilizing secondary antibody conjugation with magnetic beads, and (iv) decoding the mass‐tag that is detached from the positive resins pool by using mass spectrometric analysis. We tested a small library consisting of 27 MUC1 glycopeptides with different O‐glycosylations against anti‐MUC1 mAb clone VU‐3C6. Qualitative mass‐tag analysis showed that increasing the number of glycans leads to an increase in the binding affinity. Six glycopeptides selected from the library were validated by using a microarray‐based assay. We have developed a simple and powerful method, which is called ‘angled sample holder method’, to characterize a topographic structured sample such as microsized via hole of ball grid array using time‐of‐flight SIMS. The diameter of via holes was 100 µm and the depth was 70 µm. To remove the shaded area by incidence primary ion beam and to extract secondary ions from the bottom of a via hole, several types of angled sample holders with compensation steering plate were applied on the basis of simulation results using SIMION code. Lower respiratory tract infection is one of the most common infectious diseases. However, conventional methods for detecting infectious pathogens are time‐consuming, and generally have a limited impact on early therapeutic decisions. We previously reported a rapid and sensitive method for detecting such pathogens using stuffer‐free multiplex ligation‐dependent probe amplification coupled with high‐resolution CE‐SSCP. In this study, we report an application of this method to the detection of respiratory pathogens. As originally configured, this method was capable of simultaneously detecting seven bacterial species responsible for lower respiratory tract infections, but its detection limit and assay time were insufficient to provide useful information for early therapeutic decisions. To improve sensitivity and shorten assay time, we added a target‐specific preamplification step, improving the detection limit from 50 pg of genomic DNA to 500 fg. We further decreased time requirements by optimizing the hybridization step, enabling the entire assay to be completed within 7 h while maintaining the same detection limit. Taken together, these improvements enable the rapid detection of infectious doses of pathogens (i.e. a few dozen cells), establishing the strong potential of the refined method, particularly for aiding early treatment decisions. 相似文献

Antibody‐conjugated magnetic nanoparticles (Ab‐MNPs) have potential in pathogen detection because they allow target cells to be easily separated from complex sample matrices. However, the sensitivity and specificity of pathogen capture by Ab‐MNPs generally vary according to the types of MNPs, antibodies, and sample matrices, as well as preparation methods, including immobilization. Therefore, achieving a reproducible analysis utilizing Ab‐MNPs as a pathogen detection method requires accurate characterization of Ab‐MNP capture ability and standardization of all handling processes. In this study, we used high‐resolution CE‐single strand conformational polymorphism coupled with a stuffer‐free multiplex ligation‐dependent probe amplification system to characterize Ab‐MNPs. The capture ability of Ab‐MNPs targeting Salmonella enteritidis and nine pathogens, including S. enteritidis, was analyzed in phosphate buffer and milk. The effect of storage conditions on the stability of Ab‐MNPs was also assessed. The results showed that the stuffer‐free multiplex ligation‐dependent probe amplification system has the potential to serve as a standard characterization method for Ab‐MNPs. Moreover, the precise characterization of Ab‐MNPs facilitated robust pathogen detection in various applications. 相似文献

CE‐SSCP analysis is a well‐established DNA separation method that is based on variations in mobility caused by sequence‐induced differences in the conformation of single‐stranded DNA. The resolution of CE‐SSCP analysis was improved by using a Pluronic polymer matrix, and it has been successfully applied in various genetic analyses. Because the Pluronic polymer forms a micellar cubic structure in the capillary, it provides a stable internal structure for high‐resolution CE‐SSCP analysis. We hypothesized that formation of micellar cubic structure is influenced by the end hydroxyl group of the Pluronic polymer, which affords structural stability through hydrogen bonding. To test this hypothesis, the hydroxyl group was halogenated to eliminate the hydrogen bonding without disturbing the polarity of polymer matrix. CE‐SSCP resolution of two DNA fragments with a single base difference was significantly worse in the halogenated polymer matrices due to band broadening. The viscoelastic properties of control (which has hydroxyl group), chlorinated, and brominated F108 solution upon heating were also investigated by rheological experiments, and we found that gelation was significantly associated with resolution. In this series of experiments, the effect of the hydroxyl group in Pluronic polymer matrix on separation resolution of CE‐SSCP analysis was demonstrated. 相似文献

Accuracy, simplicity, and cost‐effectiveness are the most important criteria for a genotyping method for SNPs compatible with clinical use. One method developed for SNP genotyping, ligase‐based discrimination, is considered the simplest for clinical diagnosis. However, multiplex assays using this method are limited by the detection method. Although CE has been introduced as an alternative to error prone microarray‐based detection, the design process and multiplex assay procedure are complicated because of the DNA size‐dependent separation principle. In this study, we developed a simple and accurate multiplex genotyping method using reaction condition‐optimized ligation and high‐resolution CE‐based SSCP. With this high‐resolution CE‐SSCP system, we are able to use similar‐sized probes, thereby eliminating the complex probe design step and simplifying the optimization process. We found that this method could accurately discriminate single‐base mismatches in SNPs of the tp53 gene, used as targets for multiplex detection. 相似文献

The independent isomeric-yield ratios of 89m,gNb for the 93Nb(γ, 4n) 89m,gNb reaction with bremsstrahlung energies of 45-, 50-, 55-, 60-, and 70-MeV were measured by the activation and the off-line γ-ray spectrometric technique at 100 MeV electron linac of the Pohang accelerator laboratory. The isomeric-yield ratios of 89m,gNb for the natZr(p, xn) 89m,gNb and the 89Y(α, 4n) 89m,gNb reactions were measured by using a stacked-foil activation technique with the proton energies of 19–45 MeV and alpha energies of 38.9-, 40.5-, and 42.5-MeV at the MC-50 cyclotron of Korea Institute of Radiological and Medical Sciences. The measured isomeric-yield ratio of 89m,gNb from the 93Nb(γ, 4n), natZr(p, xn), and 89Y(α, 4n) reactions were compared with the similar literature data in the 89Y(3He, 3n) reaction. It was found that the isomeric yield ratio of 89m,gNb increases with projectile energy, which indicate the effect of excitation energy. However, for the similar compound nucleus with the same excitation energy, the isomeric-yield ratios of 89m,gNb in the 89Y(α, 4n) and 89Y(3He, 3n) reactions are higher than those in the 93Nb(γ, 4n) and natZr(p, xn) reactions, which indicates the role of input angular momentum. The isomeric-yield ratios of 89m,gNb in the 93Nb(γ, 4n), natZr(p, xn), 89Y(α, 4n), and 89Y(3He, 3n) reactions were also calculated theoretically using computer code TALYS 1.4. The theoretical isomeric-yield ratios of 89m,gNb from four reactions increase with excitation energy. However, the theoretical value are significantly higher than the experimental data in the 93Nb(γ, 4n) and natZr(p, xn) reactions but slightly lower or comparable in the 89Y(α, 4n) rand 89Y(3He, 3n) reactions. 相似文献
